Epidermal Layer or AtML1 Cell Layer

pAtML1:mGFP5-ER: Arabidopsis thaliana meristematic layer1 (AtML1) promoter driving the expression mGFP5 and marks the L1 cell layer of SAM.

Central Zone (CZ) L1 Layer

pHMG::H2B:YFP: High Mobility Group (HMG, locus At1g04880) 3kb upstream sequence including the first exon was amplified from WT-Ler and fused with H2B:YFP.  HMG mRNA is detected specifically in the L1 layer of central zone of SAM.

Sub Epidermal Layer or L2 Cell Layer

pHDG4:H2B:YFP: HOMEODOMAIN GLABROUS4 (HDG4), HDG4 promoter fused with H2B:YFP show the expression in the L2 layer of shoot apex.

Stem Cells

pCLV3::mGFP5-ER: CLAVATA3 promoter driving the expression of endoplasmic reticulum localized GFP (21). CLV3 encodes a small-secreted protein and it is expressed in stem-cells within the CZ of SAMs.

pLAS:LAS:mGFP5: LATERAL SUPPRESSOR (LAS) promoter gene fusion driving the expression of GFP.  LAS promoter gene fusion is detected in all boundaries between the organ primordia and SAM.

pKAN1:KAN1:mGFP5: KANADI1 (KAN1) promoter gene fusion driving the expression  of GFP. KANADI’s are involved in abaxial cell fate determination in the SAM.  KAN1 is expressed in a ring around organ primordia and SAM boundaries.

pCHOLINE KINASE::H2B:YFP: A 1.5kb  CHOLINE KINASE  genomic fragment above the start codon amplified from WT-Ler and cloned adjacent to H2B:YFP. In situ hybridization studies detected CHOLINE KINASE mRNA expression in the rib zone

Vasculature Cell Types

AtHB8::H2B:YFP: A 2kb Arabidopsis thaliana Homebox8 (AtHB8) promoter region was amplified from WT-Ler by PCR and cloned in front of H2B:YFP. AtHB8 expression is detected exclusively in the xylem of the root and shoot.

AtSUC2::H2B:YFP:  To label the companion cells of phloem, a 2kb genomic fragment of Arabidopsis thaliana Sucrose-proton symporter (AtSUC2) promoter was amplified from WT-Ler by PCR and cloned adjacent to H2B:YFP  To label the companion cells of phloem, a 2kb genomic fragment of Arabidopsis thaliana Sucrose-proton symporter (AtSUC2) promoter was amplified from WT-Ler by PCR and cloned adjacent to H2B:YFP.
